چکیده

Major Accomplishments to date: 1. Genetic Manipulations. The fungal transformation expression vector, pBARGPE1 was provided by Dr. Krishna Podila, Michigan Technological University. The vector had the Aspergillus nidulans gpdA promoter and the A. nidulans trpC terminator flanking a multiple cloning site. The ß-glucuronidase gene (GUS) was provided by Dr. Randy Allen, Texas Tech University. The 1.8 kb GUS gene was excised by restriction enzyme digestion of pRTL2-GUS as a BamH1-EcoR1 fragment. The BamH1 site was blunt-ended using T4 DNA polymerase. Transformation vector pBARGPE1 was digested with Xho1, blunt-ended, and digested with EcoR1. The GUS gene was thus directionally cloned under control of the gpdA promoter using T4 DNA ligase. The resulting 7.3 kb plasmid construct was transformed into Escherichia coli DH5a using CaCl2 transformation.

Studies on the Virome of the Entomopathogenic Fungus Beauveria bassiana Reveal Novel dsRNA Elements and Mild Hypervirulence

The entomopathogenic fungus Beauveria bassiana has a wide host range and is used as a biocontrol agent against arthropod pests. Mycoviruses have been described in phytopathogenic fungi while in entomopathogenic fungi their presence has been reported only rarely. Here we show that 21.3% of a collection of B. bassiana isolates sourced from worldwide locations, harbor dsRNA elements. Molecular cha...

Enzymatic comparison and mortality of Beauveria bassiana against cabbage caterpillar Pieris brassicae LINN

Beauveria bassiana, an entomopathogenic fungus, is the alternative biocontrol agent exploited against major economic crop pests. Pieris brassicae L. is an emerging pest of the Brassicaceae family. Therefore, in the present study, fungal isolates of Beauveria bassiana, viz. MTCC 2028, MTCC 4495, MTCC 6291, and NBAII-11, were evaluated for their virulence against third instar larvae of P. brassic...

Repellency of callicarpenal and intermedeol against workers of imported fire ants (Hymenoptera: Formicidae).

Callicarpenal and intermedeol are two insect-repellent terpenoids isolated from leaves of American beautyberry (Callicarpa americana L.; Verbenaceae) and Japanese beautyberry (Callicarpa japonica Thunb.). The repellency of these two terpenoids against workers of red imported fire ants, Solenopsis invicta Buren, black imported fire ants, Solenopsis richteri Forel, and a hybrid of these two speci...

Fire ant-detecting canines: a complementary method in detecting red imported fire ants.

In this investigation, detection dogs are trained and used in identifying red imported fire ants, Solenopsis invicta Buren, and their nests. The methodology could assist in reducing the frequency and scope of chemical treatments for red imported fire ant management and thus reduce labor costs and chemical use as well as improve control and quarantine efficiency. Three dogs previously trained fo...

Host Specificity and Risk Assessment of Releasing the Decapitating Fly Pseudacteon curvatus as a Classical Biocontrol Agent for Imported Fire Ants

p 1 Host specificity of the decapitating fly Pseudacteon curvatus was studied to determine whether this species is suitable for release as a classical biocontrol agent of imported fire ants in the United States. A series of no-choice tests with 19 species of ants from 12 genera showed that P. curvatus will not develop in ants outside the genus Solenopsis. P. curvatus successfully parasitized th...
